Overview of Solid Phase Extraction (SPE) Column: Solid phase extraction (SPE) is the process of using solid adsorbents to adsorb target compounds from liquid samples, separate them from the sample matrix and interfering compounds, and then elute with eluent or desorb by heating to achieve the separation and enrichment of target compounds.
It utilizes the poor adsorption ability of analytes in different media to purify the target substance, effectively separating it from interfering components, greatly enhancing the detection ability of analytes, especially trace analytes, and improving the recovery rate of the tested sample.
Since its emergence in the late 1970s, SPE technology has developed rapidly and is widely used in fields such as environment, pharmaceuticals, clinical medicine, and food. Solid phase extraction, as a sample pretreatment technique, has been increasingly widely used in laboratories.
SPE column classification:
SPE columns are classified into the following three categories based on their mechanism of action.
1. The adsorbents used in solid-phase extraction are polar, depending on the interactions between the polar functional groups of the target compound and the polar functional groups on the adsorbent surface, including hydrogen bonding, π - π bond interactions, dipole dipole interactions, dipole induced dipole interactions, and other polarity polarity interactions.
The adsorbents and target compounds used in reverse phase solid-phase extraction are usually non-polar or weakly polar, mainly relying on non-polar non-polar interactions, such as van der Waals forces or dispersion forces.
3. Ion exchange solid-phase extraction relies on the electrostatic attraction between the target compound and the adsorbent.
The operation steps vary slightly depending on the different mechanisms of filler retention (whether the filler retains the target compound or impurities). 1. The solid-phase extraction operation of retaining the target compound with fillers generally involves four steps (see Figure 1):
(1) Activation - Remove impurities from the small column and create a certain solvent environment.
(2) Sample loading - Dissolve the sample in a certain solvent, transfer it into the column, and keep the components on the column.
(3) Rinse - remove interfering substances.
(4) Elution - Use a small volume of solvent to elute the test substance and collect it.
